Reverse Complement Polymerase Chain Reaction (RC-PCR) is a high-throughput library preparation method used to analyse hypervariable areas, also known as mutation hotspots, in extremely small amounts of nuclear mitochondrial DNA (mtDNA) that is heavily degraded. This method is particularly useful for complicated materials that do not have a complete Short Tandem Repeat (STR) profile. For laboratories in countries where rules or regulations prevent whole genome sequencing in forensics.



RC-PCR enables the simultaneous occurrence of multiplex targeted amplification, sequencing indexing, and adaptor insertion in a single closed tube reaction.
